Lopinavir, an HIV-1 peptidase inhibitor, induces alteration on the lipid metabolism of Leishmania amazonensis promastigotes

The anti-leishmania effects of HIV peptidase inhibitors (PIs) have been widely reported; however, the biochemical target and mode of action are still a matter of controversy in Leishmania parasites. Considering the possibility that HIV-PIs induce lipid accumulation in Leishmania amazonensis, we analysed the effects of lopinavir on the lipid metabolism of L. amazonensis promastigotes. To this end, parasites were treated with lopinavir at different concentrations and analysed by fluorescence microscopy and spectrofluorimetry, using a fluorescent lipophilic marker. Then, the cellular ultrastructure of treated and control parasites was analysed by transmission electron microscopy (TEM), and the lipid composition was investigated by thin-layer chromatography (TLC). Finally, the sterol content was assayed by gas chromatography–mass spectrometry (GC/MS). TEM analysis revealed an increased number of lipid inclusions in lopinavir-treated cells, which was accompanied by an increase in the lipophilic content, in a dose-dependent manner. TLC and GC–MS analysis revealed a marked increase of cholesterol-esters and cholesterol. In conclusion, lopinavir-induced lipid accumulation and affected lipid composition in L. amazonensis in a concentration–response manner. These data contribute to a better understanding of the possible mechanisms of action of this HIV-PI in L. amazonensis promastigotes. The concerted action of lopinavir on this and other cellular processes, such as the direct inhibition of an aspartyl peptidase, may be responsible for the arrested development of the parasite.

Objective
To investigate the relationship between dietary patterns and self-perceived body shape silhouette and BMI in a sample of Mexican women.
DesignA cross-sectional analysis of dietary habits from baseline data of a large cohort study (EsMaestra) conducted in 2006–2008.
SettingThe state of Veracruz, Mexico.
SubjectsMexican teachers (n 20 330) provided information on body shape silhouette at baseline, changes in body shape silhouette and BMI, as well as information on sociodemographic variables and lifestyle.
ResultsThe median BMI was 26·8 kg/m2; 43 % of women were overweight and 24 % were obese. The carbohydrates, sweet drinks and refined foods pattern was associated with a greater risk of having a large silhouette and a large BMI (BMI ≥ 30·0 kg/m2v. BMI < 25·0 kg/m2; ORT1−3 = 1·86, 95 % CI 1·56, 2·22 and 1·47, 95 % CI 1·28, 1·69, respectively) with a significant trend when comparing the first and third tertiles of intake. The fruit and vegetable pattern was associated with a lower risk of having a large silhouette and a large BMI (ORT1−3 = 0·68, 95 % CI 0·57, 0·82 and ORT1−3 = 0·77, 95 % CI 0·67, 0·88, respectively) with a significant decreasing trend. Similar results were observed when change in silhouette (from 18 years of age to current silhouette) was considered.
ConclusionsHigh intakes of carbohydrates, sweet drinks and refined foods are related to larger silhouettes. Public health intervention improving access to healthy dietary guidelines, healthy food choice in the work place, promotion of physical activity and regulation of beverages with a high sugar content and of refined foods should be considered.

Cholinergic-rich grafts have been shown to be effective in restoring maze-learning deficits in rats with lesions of the forebrain cholinergic projection system. However, the relevance of those studies to developing novel therapies for Alzheimer's disease is questioned.

We present flux-calibrated integrated spectra in the optical spectral range of Galactic open clusters (GOCs) and Magellanic Cloud (MC) stellar clusters (SCs) obtained at CASLEO (Argentina). The SC parameters were derived using the equivalent-width (EW) method and the template-matching procedure by comparing the line strengths and continuum distribution of the cluster spectra with those of template spectra with known parameters. MC cluster reddening values were also estimated by interpolation between the available extinction maps. The derived ages for the GOCs range from 3 Myr to 4 Gyr, while those of the MC SCs vary from 3 Myr to 7 Gyr. E(B−V) colour-excess values in the MCs appear to be all lower than 0.17 mag, while those of the GOCs range from 0.00 to 2.40 mag. The present data led us to upgrade the spectral libraries of reference spectra or templates of solar and MC metallicities.

Summary
Introduction
In spite of early attempts at neural transplantation as long ago as the late nineteenth century, throughout most of the twentieth century it was widely believed that the mammalian brain was relatively fixed and immutable in adulthood, incompatible with receiving and supporting viable transplants. However, at the end of the 1960s, two discoveries challenged this received view: the demonstration that sprouting and reorganization of axons can indeed take place after damage in adult central nervous system (CNS) pathways; and new experimental methods for transplanting nerve cells that were remarkably successful in yielding surviving grafts.
In the first decade after these pioneering studies, attention focused on understanding the basic cellular and developmental biology of neural transplantation in a variety of model systems. Cells were transplanted into the CNS of adult rats using a wide variety of experimental model systems – anterior eye chamber, spinal cord, cerebellum, and diverse forebrain sites including cortex, hypothalamus, striatum, and hippocampus. In the first wave of studies (as illustrated in Fig. 17.1), pieces of neural tissue were implanted into natural cavities such as the anterior chamber of the eye, the brain ventricles or choroidal fissure. In the search for a greater flexibility of graft placement, other studies introduced inoculation of tissue fragments directly into brain parenchyma, although such grafts did not survive well, or the creation of artificial cavities with a rich vascular lining that would nourish newly grafted tissues.

The young stellar object SVS 13 has been proposed as the powering source of the HH 7-11 objects. Recent observations have revealed that in the radio continuum (3.6-cm) the source is a binary separated by about 0.3 in the east-west direction. We present Very Large Array observations, made in the highest angular resolution A configuration, of H2O maser emission toward this source. Our data show that the H2O spots appear to be segregated in two velocity groups: a group with radial velocity similar to that of the ambient cloud (VLSR ⋍ 8 km s−1) that is associated with the western radio source, and a blueshifted (by ∼20 km s−1) velocity group that is associated with the eastern radio source. We discuss the possible implications of this observation.
